Detecting low-frequency DNA mutations remains a daunting task due to the background noise caused by wild-type (WT) DNA. In this article, we present a recent breakthrough that simplifies DNA mutation detection by eliminating WT DNA background using phosphorothioated DNA-guided endonuclease excision.
