A new CRISPR/Cas9 system with high editing efficiency and fidelity
We identified a novel type II-A nuclease, FrCas9. It possesses comparable cutting efficiency to SpCas9 but with lower off-target effect. Furthermore, the specific 5'-NNTA-3' PAM enables FrCas9 to directly target the TATA-box to regulate gene expression.
Chemiluminescent screening of specific hybridoma cells via a proximity-rolling circle activated enzymatic switch
A 1-day homogeneous chemiluminescent screening protocol for specific hybridoma cells via a proximity-rolling circle activated enzymatic switch (P-RCAES) strategy has been developed. This CL assay enables the screening of specific hybridoma cells on conventional 96-well plate at single-cell level.
Omnidirectional-total-reflection metagratings: A compact strategy for guiding mechanical waves
Guidance of classical waves is key to many technologies, but high-efficiency, omnidirectional performance is difficult to achieve. Here, an ultrathin, broadband metagrating is proposed for suppression of parasitic diffraction and guiding mechanical waves along an arbitrary path.
